Pathway of noradrenaline formation from DOPA.

چکیده
Three compounds have each been postulated as the possible immediate precursor of noradrenaline (1) : p-hydroxyphenylethanolamine, dihydroxyphenylserine, and hydroxytyramine. Erspamer (2) found p-hydroxyphenylethanolamine in the salivary gland of the octopus; however, the compound has not been reported to be present in mammalian tissues, nor has it been shown that it can be converted to noradrenaline by enzymatic action. Beyer (3) and Blaschko et al. (1) have demonstrated that dihydroxyphenylserine is slowly decarboxylated by rabbit and guinea pig kidney extracts to form noradrenaline. Schmiterlow (4) found that the noradrenaline content of rabbit urine was increased after injection of dihydroxyphenylserine. This compound, however, has not been shown to occur in mammalian tissue. Goodall and Kirshner (5) demonstrated that adrenal slices and homogenates could form adrenaline and noradrenaline from tyrosine, from dihydroxyphenylalanine, and from hydroxytyramine, and presented evidence that, in the formation of noradrenaline, tyrosine was successively converted to dihydroxyphenylalanine, hydroxytyramine, and noradrenaline. It is now shown that the alternate pathway, tyrosine to dihydroxyphenylalanine to dihydroxyphenylserine to noradrenaline (Fig. I), is involved, if at all, to only a minor extent in the formation of noradrenaline.

The uptake of dopamine-14C and its conversion to noradrenaline-14C in rats.
The scheme of biosynthesis of noradrenaline from dopa in the mammalian tissues was presented by Blaschko (1). The conversion of dopamine to noradrenaline, mediated by dopamine-Q-hydroxylase (2), was shown to occur in the intracellular granules of the adrenal medulla (3) and heart (4, 5). The administration of tyrosine-14C (6) or dopa 14C (7 , 8) to rats results in formation of noradrenaline-14C...
متن کاملEffects of dopamine and DOPA on the isolated rabbit's atrium treated with reserpine.
In the foregoing report Matsuo and Tachi (1) showed that though the isolated atrium of rabbit which was beating in vitro took up considerable amounts of noradrenaline added to the bath fluid, the content of noradrenaline of the restarted atrium by addition of nor adrenaline did not significantly differ from the reduced content of noradrenaline of the atrium which was ceased to beat by addition ...
متن کاملThe formation of singlet oxygen during oxidation of catechol amines as detected by infrared chemiluminescence and spectrophotometric method.
1 delta g----3 sigma-g near infrared chemiluminescence from the peroxidation reaction of dopa, dopamine, noradrenaline and adrenaline was measured. The spectrophotometric method based on the bleaching of p-nitrosodimethylaniline was applied to check the generation of singlet oxygen during oxidation of the above-mentioned catecholamines.
متن کاملSeparation of adrenaline, noradrenaline, and hydroxytyramine by ion exchange chromatography.
Methods for the separation of adrenaline, noradrenaline, hydroxytyramine, and dihydroxyphenylalanine (dopa) by employing starch columns (2, 3), paper chromatography (4-7), or countercurrent extraction (8) have limited usage either because of the length of time required for the fractionation or because of the small amounts of material that can be separated. A procedure is described below in whic...
